Resumo

Recent work has suggested a subunit chromatin structure (nucleosome) in which two of each of the core histones H3, H4, H2A, and H2B are complexed with about 140 base pairs of DNA (for review, see Weintraub et al. 1976). Both histone-histone and histone-DNA interactions are responsible for the organization and maintenance of such a structure. The asymmetry in charge distribution of each of the core histones suggests that the nonstructured positively charged N termini are likely to bind to the phosphates on the DNA backbone (Bradbury and Crane-Robinson 1971; Weintraub and Van Lente 1974; Li 1975). The nonbasic portion of the molecules is probably involved in histone-histone interaction as well as in the correct folding of the DNA in the nucleosome. This possibility is suggested by the destruction in urea of the periodic structure of chromatin (Carlson et al. 1975). Studies of histones in solution have demonstrated the existence of
