and production of complement is increased following brain injury and neurodegeneration.Here, we discovered that complement receptor 2 (Cr2, also known as CD21/ CD35) is functionally expressed on neural progenitor cells (NPCs) and regulates their proliferation and responses to injury.By stereological quantification, we found that Cr2 knockout mice have significantly increased neurogenesis with more than 3-fold increase in DCX+ cells in the hippocampus of 20-months old Cr2 knockout mice.Additionally, we found that stimulation of NPCs with two ligands of Cr2, complement C3d and interferon-alpha (IFNa), reduced proliferation and neurosphere formation in vitro; this was restored by treatment with Cr2 neutralizing antibody.Corroborating these findings, we found that intracerebral injection of Cr2 ligands into the left hippocampus of wild type mice resulted in an average 35% decrease of BrdU + DCX+ cells in the ipsilateral compared with the saline injected side.Moreover, injection of C3d had no effect in Cr2 knockout mice.In conclusion, we provide evidence for a role of complement receptor 2 in regulating adult neurogenesis in the mouse hippocampus and postulate that the Cr2-mediated regulation of NPCs may mechanistically contribute to maintaining a neurogenic niche.
