ResumoRoute finder: The Passerini three-component reaction of a carboxylic acid, an aldehyde (or ketone), and an isocyanide to give an α-acyloxycarboxamide actually follows a four-component mechanism involving an additional molecule of carboxylic acid (see scheme). The discovery was made by applying a new “reaction route explorer” to the simplest Passerini reaction (HCOOH+HCHO+CH3NC).
