Simultaneous Analysis of Primidone, Phenobarbital, and Phenylethylmalonamide in Plasma and Brain

The gas-liquid chromatographic method we have developed analyzes primidone and its two active metabolites, phenobarbital and phenylethylmalonamide (PEMA), simultaneously with the use of a modified Supelco SP 2510-DA column and flame-ionization detection. Extraction and analysis are applicable to both plasma and brain samples. The drugs are quantitated underivatized. Extraction efficiency exceeded 91% for all components in both plasma and brain with coefficients of variation for primidone, phenobarbital, and PEMA of 16, 9 and 11% respectively. Mean levels for patient samples analyzed were 8.6p.g/ml for primidone, 26.0 p.g/ml for phenobarbital, and 6.2/~g/ml for PEMA. The method was used to analyze concurrent plasma and brain tissue In a kinetic study of primidone and motabolites in mice.
