SUMMARY Susceptible chicken embryos were inoculated at 7 days of incubation with dilutions of one of three strains (embryo-adapted and two field strains) of avian encephalomyelitis virus (AEV). Beginning at 3 days postinoculation (PI), thawed frozen sections of various tissues were reacted with fluorescent-antibody (FA) conjugates prepared from antiserums against the three virus strains. Specific reactions were first found 5 days PI, in brain sections from embryos infected with the egg-adapted strain. At 6 days PI, sections of heart, proventriculus, ventriculus, duodenum, and pancreas became positive. Skeletal muscle (thigh) became positive at 7 days PI, with lung and kidney following on the 8th day. No specific reactions were observed in embryonic bursa of Fabricius or spleen. No specific FA reactions were observed in any tissues from embryos infected with the two field strains of AEV. When chicks were hatched, however, from field-virus-infected embryos, specific reactions were observed in tissues 3 days posthatching.
