Learn more.Copy URL Share a linkShare onEmailFacebookTwitterLinkedInRedditWechat Abstract Summary In order to study the accuracy and the reproducibility of bovine leukocyte counts by means of electronic counters, a series of comparative tests was carried out in which 14 laboratories were involved. The essential results are as follows: 1 There were enormous differences between the leukocyte counts obtained in different laboratories which could not be reduced even after careful calibration following an agreed procedure. 2 Two main sources of this variation could be identified: (1) there was ample discretionary space for setting the threshold because most of the counters gave a wide plateau along the calibration curve, (2) the automatic diluting equipment gave rise to persistent errors in the dilution of the blood. 3 Hemocytometer counts, which were done in parallel, were in general somewhat lower than the electronic counts. Generally, the hemocytometer was superior to the electronic counter as far as reproducibility was concerned. 4 A considerable inter-laboratory variation was also observed in the determination of the percentage of lymphocytes. It is suggested that the tcchnique of preparation of the smears is responsible for this. 5 It is concluded that satisfactory standardisation of the white blood cell counting is not possible at the present time. I Technische Probleme bei der Leukozytenzählung Zusammenfassung Bovine Hämatologie Zum Studium der durch elektronische Zählgeräte ermittelten Leukozytenzahl auf Genauigkeit und Reproduzierbarkeit wurde eine Reihe von vergleichenden Untersuchungen unter Beteiligung von 14 Laboratorien durchgeführt, deren Ergebnisse wie folgt zusammengefaßt werden können: 1 Die in den verschiedenen Labors gefundenen beträchtlichen Unterschiede zwischen den ermittelten Leukozytenwerten konnten auch nach sorgfältiger Eichung der Geräte, die nach einheitlichem Schema durchgeführt worden war, nicht vermindert werden. 2 Zwei Hauptursachen für diese Unterschiede konnten identifiziert werden: Einmal bestand ein großer Spielraum für die Einstellung des Schwellen-wertes ("Threshold"), weil die Eichkurven der meisten Geräte ein ausgedehntes Plateau aufwiesen, zum anderen gingen Fehler bei den Verdünnungen auf systematische Fehler bei den Verdünnungsgeräten zurück. 3 Die Werte, die bei den parallel durchgeführten Kammerzählungen ermittelt wurden, lagen in der Regel etwas niedriger als die durch die elektronischen Geräte erzielten Werte; in Bezug auf die Reproduzierbarkeit mußte den Zählkammern jedoch der Vorzug eingeräumt werden. 4 Unterschiede bei der Ermittlung des Prozentsatzes der Lymphozytenwerte zwischen den einzelnen Labors dürften auf die Präparationstechnik und einen "Persönlichkeitsfaktor" des Untersuchenden zurückzuführen sein. 5 Es muß festgestellt werden, daß gegenwärtig eine zufriedenstellende Standardisierung bei der Bestimmung des weißen Blutbildes unmöglich ist. I Problèmes techniques lors de la numération des leucocytes Résumé Hématologie bovine Pour tester l'exactitude et la reproduction des résultats de numération des leucocytes au compteur électronique, on a comparé les données obtenues dans 14 laboratoires et on les a résumés comme suit: 1 Les différences marquées trouvées dans les différents laboratoires n'ont pu être évitées malgré l'étalonage soigneux des appareils et l'unité de méthode. 2 Deux causes principales de ces différences ont pu être identifiées: il y eut d'une part une grande variation pour la mise au point de la valeur de grossissement ("Threshold") étant donné que les courbes étalons de la plupart des appareils présentaient un plateau étendu et d'autre part les erreurs de dilutions furent imputables à des erreurs systématiques des appareils de dilution. 3 Les valeurs établies parallèlement par comptage optique furent en général plus basses que celles des appareils électroniques. La préférence devrait être accordée à la numération optique en ce qui concerne la reproductibilité. 4 Les différences dans le pourcentage des valeurs lymphocytaires devraient être attribuées à la technique de préparation et à un "facteur de personnalité" des examinateurs. 5 Il faut admettre qu'une standardisation satisfaisante de la détermination de leucocytes n'est pas possible. I Problemas técnicos en el recuento de leucocitos Resumen Hematología bovina Para el estudio de la cantidad de leucocitos determinada mediane aparatos contadores electrónicos en cuanto a exactitud y reproducibilidad, se llevó a cabo una serie de pesquisas comparativas con participación de 14 laboratorios, cuyos resultados se pueden resumir como sigue: 1 Las diferencias considerables halladas en los distintos laboratorios entre los valores leucocitarios determinados no se pudieron aminorar tampoco después de contrastar los aparatos, lo cual se hizo con arreglo a un esquema uniforme. 2 Se pudieron identificar dos causas principales que condicionan estas diferencias: Por un lado existía un espacio de huelgo grande para el ajuste del valor dintel, ya que las curvas de contraste de la mayoría de los aparatos ostentaban una meseta amplia, por otro lado se derivan los equívocos en las diluciones de errores sistemáticos en los aparatos de dilución. 3 Los valores apreciados en los recuentos de cámara, llevados a cabo de forma paralela, solían ser algo más inferiores que los valores obtenidos con ayuda de los aparatos electrónicos; sin embargo, con respecto a la reproducibilidad se debía conceder la preferencia a las cámaras de recuento. 4 Las diferencias en el cálculo del porcentaje de los valores linfocitarios entre los distintos laboratorios deberían atribuirse a la técnica preparatoria y a un "factor de personalidad" de la persona examinadora. 5 Hay que hacer constar que resulta en la actualidad imyosible una estandardización satisfactoria en la verificación del cuadro hemático blanco.
