Callus Induction and Plant Regeneration from Floral Organ of Lilium ‘Manissa'

ResumoFloral organs of Lilium ‘Manissa' were used as the explants for callus induction and plant regeneration.The results showed that several parts of floral organs,such as filament,style,ovary,petal and anther,had different re-differentiation capabilities,and the callus induction rates of them were descending in the order as before.The filament was the optimum explants for callus induction.The optimum media for callus induction,proliferation,bud re-differentiation and plantlets rooting were MS+0.5 mg/L 6-BA+0.3 mg/L NAA+0.3 mg/L 2,4-D,MS+0.5 mg/L 6-BA+0.5 mg/L NAA,MS+0.6 mg/L 6-BA+0.6 mg/L NAA and MS+0.1 mg/L NAA,respectively.The percentage of callus induction was 69.56%,regeneration frequency was 100% and rooting rate was 100%.Finally,the plantlets were transplanted onto the matrix made from peat,vermiculite,perlite,garden soil and sand in proportion as 2 ∶ 1 ∶ 1 ∶ 1 ∶ 1,and the survival rate was 92%.
