Effects of Jupi Decoction on pica and gastrointestinal hormones in rats induced by cis-platinum

ResumoObjective: To observe the influence of Jupi Decoction on pica and gastrointestinal hormones in rats induced by cis-platinum, and investigate the anti-nauseamechanism of Jupi Decoction. Methods: After adaptive feeding with kaolin, 48 rats were randomly divided into 8 groups, which included control group, model group, ondansetron group, aprepitant group, high, middle and low dosage of Jupi Decoction groups, and pure traditional Chinese medicine(TCM) treatment group, and with 6 rats in each group. The pica rat models were established by using intraperitoneal injection with cis-platinum. The influence of Jupi Decoction in kaolin and food consumptions within 0-72 h were observed, and the changes in plasma gastrointestinal hormone and substance P of central and peripheral were tested by using radioimmunoassay and immunohistochemistry methods. Results: Compared with model group, high dosage of Jupi Decoction could decrease the kaolin consumptions and increase the food consumptions of rats significantly regardless of the acute phase or delayed phase(P0.05, P0.01). High and middle dosage of Jupi Decoction could inhibit the increase of cholecystokinin(CCK), calcitonin generelated peptide(CGRP) and motilin(MTL) in plasma of rats induced by cis-platinum(P0.01). High dosage of Jupi Decoction could decrease the expression of SP in AP area and antrum of stomach of rats(P0.01). Conclusion: Jupi Decoction could anti emetic caused by cis-platinum and promote gastric emptying, and the mechanism might relate with the regulating effects of Jupi Decoction on the secretion of gastrointestinal hormones and expression of SP.
