Human prolactin has been purified from pregnancy urine by hydrophobic interaction chromatography and molecular sieve chromatography. The resulting product obtained in the equivalent of 3 ug of pituitary prolactin per litre urine was analyzed by column electrophoresis in agarose suspension in combination with radioimmunoassay. At acidic pH the urinary prolactin appeared as a single component and comigrated with a pituitary reference. The pattern obtained upon electrophoresis at alkaline pH indicated the presence of seven or eight immunoactive components. By comparing this pattern with the pituitary preparation consisting of three components, it was found that the additional urinary isohormones were more acidic and their appearance was suggested to be caused by deamidation of the hormone.
