DNA-B and satellite DNA associated with PaLCuGdV has not been found. PaLCuGdV has been reported that causes leaves to curl in papaya and passionfruit (Varun et al. 2017). In 2015, PaLCuGdV and Euphorbia leaf curl virus (EuLCV) were observed in greenhouse of passionfruit plants in Goheung, South Korea (Choi et al. 2016). Additionally, in July 2015, symptoms of mottle and crinkle were observed on leaves of bell pepper (Capsicum annuum L. 'Asia Green Pimang') in a greenhouse located near a passionfruit farm in Goheung, South Korea. Nucleic acids were extracted from leaves of six bell pepper samples using the viral DNA/RNA extraction kit (iNtRON biotech, Seongnam, Korea). PaLCuGdV-specific primer sets were designed to amplify a portion of the V1 gene (PaLCuGdV-F 5′-CGTTTGGTACTCCCCAAGA-3′ and PaLCuGdV-R 5′-CAACAGCAAAGCATTCTCAGT-3′, expected amplicon size 254 bp). PCR primer sets EuLCV F/R (5′-AGTGGTCCCCCCTCCMCTAAC-3′ and 5′-CAGCCTCCGTCGAACCTTCG-3′) were designed by amplifying a 342-bp fragment for the V2 gene of EuLCV (Kil et al. 2016). Two of the bell pepper samples were found to be infected with PaLCuGdV, and both PaLCuGdV and EuLCV were detected in one sample. Rolling-circle amplification (RCA) was performed to analyze potential begomoviruses and amplify the full genome of an isolate of PaLCuGdV from DNA of single infected plant. The RCA amplified product was digested with restriction enzyme BamH I (Kil et al. 2014). The product was analyzed by electrophoresis in 1% agarose gel and found to have a fragment size of approximately 2.7 kb, which was purified and cloned into the RBC T&A vector (Real Biotech Corporation, Taiwan). Three clones were sequenced in both directions by a commercial sequencing service (Geno Tech Corp., Daejeon, Korea). The complete DNA sequences were determined to 2,732 nucleotides (GenBank accession no. KY884675, PaLCuGdV-BP6). PaLCuGdV-BP6 isolate had a genome organization of begomovirus DNA-A. BLAST search results showed that the isolate had 99.6 to 99.8% nucleotide sequence identity with PaLCuGdV-PF-1 (KC161184, passionfruit, Taiwan) and PaLCuGdV-BXG4 (KP876482, passionfruit, Taiwan), and 99.5 identity with a papaya leaf curl virus (PaLCuV) isolate from papaya in Korea (KT266873). In contrast, PaLCuGdV-BP6 shared 72.8 to 77.4% sequence identity with PaLCuV isolate Wellington (KX302713, papaya, India) and PaLCuV isolate 249-1ISBD (KY978407, chili, Pakistan). Phylogenetic analysis by the maximum likelihood method using MEGA7 revealed that the PaLCuGdV-BP6 isolate is closely related to PaLCuGdV isolates from Taiwan and also demonstrated that the PaLCuV Korea isolate (KT266873) (Byun et al. 2016) is actually an isolate of PaLCuGdV based on sequence identity. This is the first report of PaLCuGdV infecting bell pepper in the world. For the last 4 years, production of passionfruit and papaya have been rapidly increasing, with many seeds and seedlings imported from outside Korea. Other than papaya, passionfruit, and now bell pepper, no other plants have been reported as hosts of PaLCuGdV in Korea. The results of this study suggest that Korea should consider development of a virus-free seedling program to ensure that passionfruit seedlings produced domestically or imported do not carry the risk of new and emerging viruses that could impact production of bell pepper and other crops in Korea.References:Byun, H.-S., et al. 2016.
