ResumoIn this chapter, I shall review our current knowledge of the mechanism of mammary oncogenesis by the Mouse Mammary Tumor Virus (MMTV), with particular emphasis on the activation of cellular oncogenes by insertion of MMTV proviral DNA. The properties of these newly discovered cellular oncogenes will be described in some detail, and finally, I shall speculate a little on the mechanism of action of these genes. As an introduction, some of the relevant earlier discoveries on MMTV will be summarized to provide a framework of the biology of viral mammary tumorigenesis. More encyclopedic overviews of the biology and molecular biology of MMTV are found in references 1, 2 and 3.
