ResumoThe work described in this chapter resulted largely from the pioneering efforts of Lardy and his coworkers in their search for "toxic" antibiotics which inhibit respiration. The results of such studies have proved that this type of approach could be very fruitful, and much insight has been gained into the mechanism of oxidative phosphorylation and associated phenomena such as mitochondrial swelling and ion transport across membranes. In fact, the use of certain of the antibiotics has provided information which would have been difficult or perhaps impossible to obtain by any other means.
